为了解不同地区淡色库蚊样本乙酰胆碱酯酶Ι基因(ace1)多态性,探讨单链构象多态性分析(SS-CP)技术在蚊虫抗性分子监测中应用的可行性,本文采集浙江省杭州、舟山、湖州、金华等市区淡色库蚊样本,对其进行ace1基因PCR扩增及SSCP分析。结果显示,浙江省淡色库蚊现场品系以及敏感品系ace1基因片段共存在2个SSCP型,分别命名为ace1-A型(杭州、舟山、湖州现场品系以及敏感品系)和ace1-B型(金华现场品系)。经序列测定,SSCP型ace1-A和ace1-B之间存在两个碱基的差异。实验证明PCR-SSCP可快速高效分析淡色库蚊ace1基因的多态性,适于蚊虫抗性分子监测使用。
In order to understand the polymorphism of acetylcholinesterase I gene (ace1) in Culex pipiens pallens from different areas and to explore the feasibility of single-strand conformation polymorphism analysis (SS-CP) in mosquito resistance molecular monitoring, this paper collected Zhejiang province Hangzhou, Zhoushan, Huzhou, Jinhua and other urban samples of Culex pipiens pallens, their ace1 gene PCR amplification and SSCP analysis. The results showed that there were two SSCP genotypes in field strains and ace1 gene segments of Culex pipiens pallens in Zhejiang Province, which were named as ace1-A type (Hangzhou, Zhoushan and Huzhou in-situ and sensitive strains) and ace1-B strain). After sequencing, two base differences exist between SSCP type ace1-A and ace1-B. Experiments show that PCR-SSCP can quickly and efficiently analyze the Culex pipiens ace1 gene polymorphism, suitable for mosquito resistance molecular monitoring.
