目的探讨一种快速、简便、经济的骨质疏松症(osteoporosis,OP)体外模型制备方法。方法 18个月健康雌性山羊80只,取双侧新鲜股骨80对,根据脱钙时间不同,随机分为4组,每组20对:对照组(A组)、1~5 d组(B组)、6~10 d组(C组)和11~15 d组(D组)。B、C、D组股骨标本用18%EDTA脱钙液浸泡,A组不作处理。每组于对应浸泡时间段内每天取4对股骨,采用定量CT对股骨内、外侧髁进行扫描并计算骨密度(bone mineral density,BMD);采用电子万能试验机进行三点弯曲试验和拉伸、压缩极限强度试验,测量相应力学参数。结果随脱钙时间延长,A、B、C、D组股骨内、外侧髁BMD均呈下降趋势,组间比较差异均有统计学意义(P<0.05);各组股骨外侧髁BMD均显著高于股骨内侧髁,差异有统计学意义(P<0.05)。三点弯曲试验显示,A、B组破坏荷载、极限强度及弹性模量均显著高于C、D组(P<0.05);A、B组间及C、D组间比较,差异均无统计学意义(P>0.05)。压缩和拉伸极限强度试验显示,A组压缩和拉伸极限强度显著高于C、D组(P<0.05),与B组比较差异无统计学意义(P>0.05);B组两指标显著高于D组(P<0.05),与C组比较差异无统计学意义(P>0.05);C、D组间差异无统计学意义(P>0.05)。结论以18%EDTA浸泡山羊股骨6~15 d是一种快速、简便、经济的OP体外模型制备方法。
Objective To explore a rapid, simple and economical method for the preparation of osteoporosis (OP) model in vitro. Methods Eighty healthy female goats aged 18 months were randomly divided into four groups according to different time of decalcification: 80 pairs of control group (group A), 1-5 days group B (group B) ), 6 ~ 10 d group (group C) and 11 ~ 15 d group (group D). B, C, D femur specimens were decalcified with 18% EDTA soaking, A group without treatment. Each group received 4 pairs of femurs daily for the soaking time period. Quantitative CT was used to scan the medial and lateral femoral condyle to calculate the bone mineral density (BMD). Three-point bending test and stretching , Compression ultimate strength test, measuring the corresponding mechanical parameters. Results With the time of decalcification, the BMD of the femoral condyle and lateral femur of groups A, B, C and D showed a decreasing trend with significant differences between the two groups (P <0.05). The BMD of the lateral femoral condyle in each group was significantly higher In the medial femoral condyle, the difference was statistically significant (P <0.05). Three-point bending test showed that the breaking load, ultimate strength and elastic modulus of group A and group B were significantly higher than those of group C and D (P <0.05). There was no statistical difference between groups A and B and group C and D Significance (P> 0.05). Compression and tensile ultimate strength tests showed that the ultimate compressive strength and tensile strength of group A were significantly higher than those of group C and D (P <0.05), but there was no significant difference between group A and group B (P> 0.05) (P <0.05). There was no significant difference between group C and group D (P> 0.05). There was no significant difference between group C and group D (P> 0.05). Conclusion The soaking of goat femur with 18% EDTA for 6-15 days is a rapid, simple and economical method for the preparation of OP model in vitro.
