本文报告从150g人心肌中得到电泳纯和免疫纯的肌酸激酶MB和MM同工酶(OK-MB和CK-MM)5.1mg和61mg,比活性分别是1232u/mg蛋白和408u/mg蛋白。同时,还研究了CK-MB和CK-MM的部分理化性质和免疫学性质。此外,制备成功CK-MB和CK-MM抗血清。经联结标记法标记~(125)I,比放射性CK-MB约11.1×10~6Bq/μg,CK-MM约5.92×10~6Bq/μg,50%结合率的CK-MB抗血清稀释度1:75000,CK-MM1:15000,亲和常数(K值)CK-MB2.7×10~(11)L/mol,CK-MM3.1×10~9L/mol,均具有理想的抗体特异性。适用于CK-MB和CK-MM的放射免疫分析(RIA)。
