目的规范安徽茯苓菌种的分离、接种以及制备的关键技术,建立茯苓菌丝的薄层鉴别方法 ,为茯苓菌丝的成分分析进行初探。方法精选茯苓传统产区的优质茯苓菌核,进行种苓的母种、原种、栽培种的分离和扩大培养,对茯苓菌丝进行薄层鉴别。结果经分离得到的茯苓原种、栽培种菌丝生长旺盛,色白、均匀、致密、粗壮,茯苓特异香气浓郁。经TLC检视出茯苓菌丝特征性斑点。结论建立的薄层鉴别方法在一定程度上确保了茯苓菌种的质量。
Objective To standardize the separation, inoculation and preparation of key technologies of Poria cocos in Anhui Province, and to establish a thin layer identification method of Poria cocos mycelium for preliminary analysis of the composition of Poria cocos mycelium. Methods Selected high quality Poria cocos of Fuling traditional producing areas, the separation, expansion and cultivation of the mother species, original species and cultivated species of Cervus colchicus were carried out. The mycelia of Poria cocos were identified by TLC. Results The Poria cocos isolates and cultivated mycelium isolated from the cultivated species had vigorous growth, color white, uniform, dense, stout and rich in Poria. The tocomerosin was observed by TLC characteristic spots. Conclusion The established TLC method to a certain extent, to ensure the quality of Poria species.
