目的分析槲皮素对IL-6、细胞间黏附分子-1(ICAM-1)以及核转录因子-κB p65(NF-κB p65)蛋白表达的影响以探讨其对血管内皮功能的作用。方法将培养的人脐静脉血管内皮细胞(HUVEC)分为空白对照组(Control组)、TNF-α组(含10 ng/ml TNF-α)、Q10组(TNF-α+10μmol/L槲皮素)、Q20组(TNF-α+20μmol/L槲皮素)及Q50组(TNF-α+50μmol/L槲皮素)。采用MTT法检测各组细胞活力。采用ELISA法检测培养液中IL-6、ICAM-1的蛋白含量,采用蛋白免疫印迹法检测细胞NF-κB p65蛋白的表达水平。结果 Control组、TNF-α组及不同浓度的槲皮素组细胞活力比较差异无统计学意义(P>0.05)。与Control组比较,TNF-α组ICAM-1、IL-6蛋白含量均较高(P均<0.01);与TNF-α组相比,不同浓度的槲皮素组ICAM-1、IL-6蛋白含量均下降(P均<0.05),且随着槲皮素浓度的增加,其对ICAM-1、IL-6蛋白表达的抑制作用更强。TNF-α组NF-κB p65蛋白表达水平较Control组高(P<0.01);与TNF-α组相比,不同浓度的槲皮素组NF-κB p65蛋白的表达均受抑制(P均<0.05),且随着槲皮素浓度的增加,其对NF-κB p65蛋白表达的抑制作用更强。结论槲皮素对ICAM-1、IL-6及NF-κB p65均有抑制作用,其可能通过抑制NF-κB p65蛋白的表达降低内皮细胞分泌IL-6、ICAM-1水平,从而实现对血管内皮功能的保护作用。
Objective To investigate the effect of quercetin on the expression of IL-6, ICAM-1 and NF-κB p65 protein in vascular endothelial cells in order to investigate the effect of quercetin on vascular endothelial function. Methods Human umbilical vein endothelial cells (HUVECs) were divided into control group, TNF-α group (containing 10 ng / ml TNF-α), Q10 group (TNF-α + 10 μmol / Q20 group (TNF-α + 20μmol / L quercetin) and Q50 group (TNF-α + 50μmol / L quercetin). Cell viability was detected by MTT assay. The protein levels of IL-6 and ICAM-1 in culture medium were detected by ELISA, and the expression of NF-κB p65 protein was detected by Western blotting. Results There was no significant difference in cell viability between Control group, TNF-α group and quercetin group (P> 0.05). Compared with the control group, the levels of ICAM-1 and IL-6 in the TNF-α group were significantly higher than those in the control group (P <0.01). Compared with the TNF-α group, ICAM-1 and IL- (All P <0.05). With the increase of quercetin concentration, the inhibitory effect on ICAM-1 and IL-6 protein expression was stronger. The expression of NF-κB p65 in TNF-α group was higher than that in Control group (P <0.01). Compared with TNF-α group, the expression of NF-κB p65 protein in quercetin group was inhibited 0.05). With the increase of quercetin concentration, the inhibitory effect on NF-κB p65 protein expression was stronger. Conclusions Quercetin can inhibit the expression of ICAM-1, IL-6 and NF-κB p65, which may decrease the level of IL-6 and ICAM-1 secreted by endothelial cells by inhibiting the expression of NF-κB p65 protein, Endothelial function of the protective effect.
