目的:观察重组人睫状神经营养因子对在体和离体大鼠成肌细胞的生物学作用。方法:通过混合酶消化和差速贴壁分离纯化大鼠成肌细胞,用含有不同浓度的重组人睫状神经营养因子(rhCNTF)的分化液培养,观察成肌细胞增殖分化的改变情况。另外,采用胫前肌肌内注射 rhCNTF,随后利用免疫组化的方法观察在体大鼠成肌细胞的变化情况。结果:2.5~10ng/ml rhCNTF 能显著抑制亚融合成肌细胞的体外分化(P<0.01),这种抑制作用呈量效依赖性,且其作用是可逆的。rhCNTF 对在体成肌细胞前体肌卫星细胞的影响不明显,但能够使肌纤维较对照组增粗。结论:在体和离体大鼠成肌细胞对重组人睫状神经营养因子的反应性存在差异。
Objective: To observe the biological effects of recombinant human ciliary neurotrophic factor on myoblasts in vitro and in vivo. Methods: Myoblasts were purified by mixed enzyme digestion and differential adherent separation. Cultured myoblasts were cultured in different concentrations of recombinant human ciliary neurotrophic factor (rhCNTF) to observe the proliferation and differentiation of myoblasts. In addition, rhCNTF was injected into the anterior tibial muscle by intramuscular injection, and the changes of myoblasts in vivo were observed by immunohistochemistry. Results: 2.5 ~ 10 ng / ml rhCNTF could significantly inhibit the differentiation of sub-fused myoblasts in vitro (P <0.01). The inhibitory effect was dose-dependent and its effect was reversible. rhCNTF had no obvious effect on muscle cells of myoblast precursors, but could make myofibers thicker than that of the control group. Conclusions: Myoblast reactivity to recombinant human ciliary neurotrophic factor is different between in vivo and in vitro rat myoblasts.
