目的研究蓝莓改善阿尔茨海默病(AD)过程中抗氧化能力的效用。方法采用APP/PS1双转基因小鼠作为AD模型动物,分别设置AD组、AD+BB组以及对照组(CT组),每组10只小鼠。分别进行相应的饲喂蓝莓提取物或者生理盐水干预,持续喂养16 w。期间每周记录小鼠体质量及食物消耗情况,干预结束后检测大脑皮层组织的形态学改变,测定脑组织、肝脏、肾脏及血清中SOD、GSH-Px活力以及GSH、MDA水平。结果干预期间,三组小鼠的食物消耗量无明显区别,但接受蓝莓提取物干预的AD小鼠体质量有明显增加。形态学检测显示,AD小鼠大脑皮层中出现明显的神经元丢失,而接受蓝莓提取物干预的AD小鼠皮层神经元丢失有明显改善。生化检测结果显示,AD小鼠脑组织、肝脏及血清中SOD、GSH-Px活力及GSH浓度显著降低,而MDA水平则明显增高。蓝莓提取物干预组的检测结果则显示脑组织中SOD活力有显著回升,MDA下降明显,肝脏及血清中GSH-Px活力及GSH水平也明显增高,MDA水平显著降低,但肾脏中相应检测指标无明显变化。结论:AD过程中脑组织对于氧化应激非常敏感,蓝莓提取物具有较好的预防AD损伤、减轻氧化损伤的效用。
Objective To study the effect of blueberries on the anti-oxidative capacity of Alzheimer’s disease (AD). METHODS: APP / PS1 double transgenic mice were used as AD model animals. AD, AD + BB and control groups (CT) were set up, with 10 mice in each group. Respectively fed blueberry extract or saline intervention, continuous feeding 16 w. The body weight and food consumption of mice were recorded weekly. Morphological changes of cerebral cortex were detected after intervention. The activities of SOD, GSH-Px, GSH and MDA in brain, liver, kidney and serum were measured. Results There was no significant difference in food consumption between the three groups during the intervention, but the body mass of AD mice receiving blueberry extract was significantly increased. Morphological examination revealed significant neuronal loss in the cerebral cortex of AD mice, while loss of cortical neurons in AD mice receiving blueberry extract was significantly improved. Biochemical tests showed that the activity of SOD, GSH-Px and the concentration of GSH in brain, liver and serum of AD mice were significantly decreased, while MDA levels were significantly increased. The results of blueberry extract intervention group showed that the activity of SOD in brain tissue was significantly increased, MDA decreased significantly, liver and serum GSH-Px activity and GSH levels were significantly increased, MDA levels were significantly lower, but the corresponding indicators in the kidney no obvious change. CONCLUSION: Brain tissue is very sensitive to oxidative stress during AD process. Blueberry extract has good effect on preventing AD injury and reducing oxidative damage.
