目的探讨天门冬水提液对D-半乳糖致小鼠衰老过程中脏器显微结构改变的影响。方法采用D-半乳糖诱导小鼠衰老,60只小鼠随机分为6组,分别为生理盐水对照组、衰老模型组、Vc对照组、天门冬水提液高、中、低剂量组,分别皮下注射或灌胃,给药30 d后,取其心、肝、脾、肺、肾、胰、脑等主要脏器石蜡切片,苏木素-伊红(HE)染色观察显微结构。结果正常对照组和Vc组器官形态正常,结构清晰;模型组脏器均出现不同程度的损伤;天门冬给药组的小鼠,随给药浓度的增加,心肌纤维结构由模糊到清晰,由扭曲缩短变成舒展;脾脏淋巴细胞数量显著增加,排列紧密;肺泡隔逐渐恢复正常,肺泡囊结构逐渐完整;胰岛细胞数量由少到多,结构渐清晰;肝、肾细胞结构逐渐恢复正常;脑部海马CA3区神经细胞逐渐增加。结论天门冬水提液在D-半乳糖诱导小鼠衰老过程中对内脏器官损伤程度均有一定抑制作用,且随给药剂量增加抑制作用增强,但不同器官抑制效果不同,效果最明显的是脾和肺,其次是心和胰。
Objective To investigate the effects of aqueous extract of Astrus on the changes of the organ microstructure of aging mice induced by D-galactose. Methods Senescence was induced in mice by D-galactose. Sixty mice were randomly divided into 6 groups: saline control group, aging model group, Vc control group, asparagus water extract high, medium and low dose groups Subcutaneous injection or intragastric administration, after administration for 30 days, the heart, liver, spleen, lung, kidney, pancreas, brain and other major organs of paraffin sections, hematoxylin-eosin (HE) staining microstructure. Results The organs of normal control group and Vc group were normal in morphology and the structure was clear. The organs of model group showed different degrees of damage. As the concentration of drug was increased, the structure of cardiac muscle fiber changed from fuzzy to clear. The number of spleen lymphocytes increased significantly and arranged closely; the alveolar septum gradually returned to normal and the structure of alveolar sac gradually became intact; the number of islet cells gradually decreased from small to large, the structure gradually became clear; the structure of liver and kidney cells gradually returned to normal; Department of hippocampal CA3 area neurons gradually increased. CONCLUSION Astronauticus aqueous extract could inhibit the injury of visceral organs in D-galactose-induced mice senescence, and the inhibitory effect was enhanced with the increase of dosage, but the effect was different in different organs, the most obvious effect was Spleen and lungs, followed by heart and pancreas.
