目的研究极低频率电磁场(ELF)对肝癌细胞死亡结构域沉默子(SODD)和凋亡抑制基因Survivin基因表达的影响,探索ELF在肝癌治疗中的作用。方法应用RT-PCR和流式细胞仪(FCM)检测经ELF处理的肝癌细胞系BEL-7402的SODD和Survivin基因表达情况,并以L-02肝细胞为正常对照,分析ELF对肝癌细胞SODD和Sur-vivin基因表达的影响。结果经ELF处理后,BEL-7402细胞SODD和Survivin基因表达均明显下降(P<0.05),而对L-02细胞无影响(均P>0.05)。结论 ELF能促进肝癌细胞凋亡,为临床应用ELF治疗肝癌提供了线索。
Objective To investigate the effect of ELF on the expression of survivin gene and the gene silence of apoptosis in hepatocellular carcinoma (HCC), and to explore the role of ELF in the treatment of hepatocellular carcinoma. Methods The expression of SODD and Survivin in ELF-treated hepatocellular carcinoma cell line BEL-7402 was detected by RT-PCR and flow cytometry (FCM). L-02 hepatocytes were used as normal control to analyze the effect of ELF on the expression of SODD, Sur-vivin gene expression. Results After ELF treatment, the expression of SODD and Survivin genes in BEL-7402 cells were significantly decreased (P <0.05), but not in L-02 cells (all P> 0.05). Conclusion ELF can promote the apoptosis of hepatocellular carcinoma cells and provide clues for the clinical application of ELF in the treatment of liver cancer.
