目的:研究FHL1(four-and-a-half LIM domain 1)在香烟烟雾提取物(CSE)刺激的大鼠远端肺动脉平滑肌细胞(PASMCs)增殖及迁移中的作用。方法:原代培养PASMCs,分别予CSE及FHL1 siRNA转染干预,分为6组:空白组、阴性转染组、FHL1 siRNA转染组、CSE组、CSE+阴性转染组和CSE+FHL1 siRNA转染组。Real-time PCR法检测FHL1 mRNA的表达,Western blotting法检测FHL1蛋白,CCK-8法检测细胞增殖,Transwell法测定细胞迁移。结果:CSE刺激导致PASMCs增殖、迁移及FHL1蛋白表达增加(P<0.01),但FHL1 mRNA的表达无明显改变(P>0.05)。FHL1 siRNA转染PASMCs,可降低CSE所致的PASMCs增殖及迁移(P<0.01)。结论:CSE可促进PASMCs增殖与迁移以及FHL1蛋白的表达,抑制FHL1蛋白的表达可减弱CSE对PASMCs增殖和迁移的作用。这些结果表明CSE促进PASMCs增殖和迁移与FHL1蛋白有关。
AIM: To investigate the role of FHL1 in the proliferation and migration of rat distal pulmonary artery smooth muscle cells (PASMCs) stimulated by cigarette smoke extract (CSE). Methods: Primary cultured PASMCs were transfected with CSE and FHL1 siRNA respectively and divided into 6 groups: blank group, negative transfected group, FHL1 siRNA transfected group, CSE group, CSE + negative transfected group and CSE + FHL1 siRNA transfected group Dyeing group. FHL1 mRNA expression was detected by Real-time PCR, FHL1 protein by Western blotting, cell proliferation by CCK-8 assay and cell migration by Transwell assay. Results: CSE stimulated the proliferation and migration of PASMCs and increased the expression of FHL1 protein (P <0.01), but the expression of FHL1 mRNA did not change significantly (P> 0.05). FHL1 siRNA transfected PASMCs could reduce the proliferation and migration of PASMCs induced by CSE (P <0.01). CONCLUSION: CSE can promote the proliferation and migration of PASMCs and the expression of FHL1 protein. Inhibiting the expression of FHL1 protein may reduce the effect of CSE on the proliferation and migration of PASMCs. These results indicate that CSE promotes the proliferation and migration of PASMCs in relation to the FHL1 protein.
