目的探讨乳腺肿瘤组织中金属蛋白酶组织抑制物TIMP┐3及TIMP┐4mRNA表达与乳腺癌转移的关系。方法以原位杂交方法检测二甲基苯蒽(DMBA)诱发的大鼠乳腺原发瘤组织中此两种mRNA的表达水平,并观察转移组、非转移组、良性肿瘤组的表达意义。结果TIMP┐3及TIMP┐4mRNA均表达于肿瘤间质,乳腺癌非转移组表达水平显著高于转移组及良性肿瘤组。结论TIMP┐3及TIMP┐4mRNA由间质细胞产生,过度表达反映了宿主细胞对肿瘤侵袭的一种应答反应,可能是转移行为的负性调节因子。
Objective To investigate the relationship between the expression of TIMP┐3 and TIMP┐4 mRNA and the metastasis of breast cancer in breast tumors. Methods In situ hybridization was used to detect the expression levels of these two mRNAs in rat mammary gland primary tumors induced by dimethyl benzoquinone (DMBA), and the significance of expression in the metastatic, non-metastatic and benign tumor groups was observed. Results TIMP┐3 and TIMP┐4 mRNA were all expressed in tumor stroma. The expression level of non-metastasis breast cancer group was significantly higher than that of metastatic group and benign tumor group. Conclusion TIMP┐3 and TIMP┐4 mRNA are produced by mesenchymal cells. Overexpression reflects a host cell response to tumor invasion and may be a negative regulator of metastatic behavior.
