用52种RAPD引物对橡胶树11个抗白粉病品系和11个感病品系基因组DNA进行RAPD分析,找到了一个和橡胶树抗白粉病表型密切相关的DNA片段,此片段长390bp,命名为opv—390。opv—390为11个抗性品系所共有和11个感病品系所没有。用ECL(enhanced cheminesence)酶标法标记抗性品系RRIC52(CK)的OPV390为探针,经Southern blot表明,该序列在6个抗性品系中均为单一拷贝或低拷贝,而感病品系皆不含此序列,由此看来,OPV—390可作为橡胶树抗白粉病基因紧密连锁的RAPD标记。这为今后钓取橡胶树抗白粉病基因的工作打下了基础。
RAPD analysis of the genomic DNA of 11 powdery mildew resistant strains and 11 susceptible strains of rubber trees with 52 RAPD primers revealed a DNA fragment closely related to the phenotype of rubber tree powdery mildew resistance. The fragment was 390 bp in length and named opv- 390. opv-390 was shared by 11 resistant lines and 11 susceptible lines did not. The OPV390 marker with the resistant chemotype RRIC52 (CK) labeled with ECL (enhanced cheminesence) was used as a probe. Southern blot showed that the sequence was single copy or low copy in the six resistant lines, while the susceptible lines Excluding this sequence, it appears that OPV-390 can be used as a RAPD marker for the close linkage of rubber tree powdery mildew resistance genes.
