目的调查潍坊市奎文区成年居民H型高血压检出比例、血浆同型半胱氨酸(Hcy)水平,探讨血浆同型半胱氨酸关键代谢酶亚甲基四氢叶酸还原酶(MTHFR C677T)基因多态性与H型高血压的关系,为制定有效的高血压防治措施提供科学依据。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLR)方法,对潍坊市奎文区740例成年人进行MTHFR C677T基因型分型和血浆Hcy水平检测。结果调查人群H型高血压检出比例为16.35%,男性高于女性,差异具有统计学意义(P<0.05);所调查的人群Hcy水平为13.58±9.02,男性高于女性,差异具有统计学意义(P<0.05);男性、女性MTHFR C677T基因TT、CT、CC基因频率分布,差异具有统计学意义(P<0.05);H型高血压人群MTHFR C677T基因CC+CT和TT基因型频率与正常血压、非H型高血压人群比较,差异具有统计学意义(P<0.05)。结论奎文区居民的H型高血压检出比例、血浆Hcy水平均偏高且存在性别差异,MTHFR C677T基因纯合突变可能是引起Hcy水平升高,进而引发H型高血压的重要遗传因素。
Objective To investigate the detection rate of Hypertension and plasma homocysteine (Hcy) in adult residents in Kuiwen District of Weifang City, and to investigate the relationship between plasma homocysteine and MTHFR C677T, a key metabolic enzyme of plasma homocysteine, Genetic polymorphisms and H-type hypertension, to provide an effective basis for prevention and treatment of hypertension provide a scientific basis. Methods MTHFR C677T genotyping and plasma Hcy levels were detected in 740 adults in Kuiwen District, Weifang City by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLR). Results The prevalence of Hypertension in the surveyed population was 16.35%, which was higher in males than in females (P <0.05). The Hcy level in the surveyed population was 13.58 ± 9.02, which was higher in males than in females (P <0.05). The frequencies of TT, CT and CC genes in MTHFR C677T gene in males and females were significantly different (P <0.05). The frequencies of MTHFR C677T gene CC + CT and TT genotypes in H and H The difference was statistically significant between normal blood pressure group and non Hypertensive group (P <0.05). Conclusions The detection rate of Hypertension and plasma Hcy in Kuiwen district residents are both high and there are gender differences. Homozygous mutation of MTHFR C677T gene may be the important genetic factor that causes Hcy level and then Hypertension.
