目的:观察蒺藜提取物(NO.JL201)对自发性2型糖尿病模型KKAy小鼠空腹血糖(FPG)、血胰岛素(Ins)、TNF-α、IFN-β含量及其胰岛细胞TLR3、TLR4mRNA含量的影响,初步探讨其可能降糖机制。方法:KKAy小鼠随机分为模型组、JL201大(20mg/kg BW)、中(10mg/kg BW)、小(5mg/kg BW)剂量组和阳性药二甲双胍组(400mg/kg BW)。每周测定体质量,食、水量和FPG。给药4周后取血,测定小鼠FPG和Ins,计算小鼠Ins敏感性,Real time-PCR法测定胰岛细胞TLR2、TLR4 mRNA含量。结果:JL201不同剂量不同程度地减少了小鼠的进食、饮水量,明显降低KKAy小鼠的FPG(P<0.05或P<0.01)、降低血清Ins含量(P<0.05或P<0.01),明显升高KKAy小鼠胰岛素敏感性(P<0.05),减少TNF-α、IFN-β的血中分泌量。JL201同时可降低胰岛细胞TLR2、TLR4mRNA含量。结论:JL201有较好的降血糖、增强胰岛素敏感性等作用,并能明显改善多饮、多食的临床症状,其作用可能与降低胰岛细胞TLR2、TLR4mRNA含量、抑制炎症有关。
OBJECTIVE: To observe the effects of Tribulus terrestris extract (NO.JL201) on fasting plasma glucose (FPG), insulin, TNF-α, IFN-β and TLR3, TLR4 mRNA in KKAy mice with spontaneous type 2 diabetes mellitus Influence, preliminary discussion of its possible hypoglycemic mechanism. Methods: KKAy mice were randomly divided into model group, JL201 large (20mg / kg BW), medium (10mg / kg BW), small (5mg / kg BW) dose group and the positive drug metformin group (400mg / kg BW). Body weight, food, water, and FPG were measured weekly. After 4 weeks of administration, blood samples were taken for determination of FPG and Ins in mice. Ins sensitivity of mice was calculated. Real-time PCR was used to determine the content of TLR2 and TLR4 mRNA in islet cells. Results: Different doses of JL201 reduced the intake and water intake of mice, decreased the FPG of KKAy mice (P <0.05 or P <0.01), decreased the level of serum Ins (P <0.05 or P <0.01) Increase insulin sensitivity in KKAy mice (P <0.05), and decrease the amount of TNF-α and IFN-β in blood. JL201 at the same time can reduce islet cell TLR2, TLR4 mRNA content. Conclusion: JL201 has better hypoglycemic effect, insulin sensitivity and other effects, and can significantly improve the symptoms of polydipsia, polyphagia, which may be related to the reduction of islet cells TLR2, TLR4 mRNA content, inhibition of inflammation.
