目的探讨CD44及其配体骨调素(OPN)在实验性新月体肾炎发展过程中的表达和作用。方法应用兔抗鼠肾毒性血清制作新月体肾炎模型,免疫组化双标记及原位杂交技术检测CD44、OPN及其它指标。结果CD44和OPN在新月体肾炎的发展过程中表达明显升高,并和巨噬细胞在肾脏组织的局部浸润、新月体的形成及肾功能的损害密切相关。结论在实验性新月体肾炎模型,CD44和OPN在肾小球实质细胞和小管间质细胞显著表达,并可能在新月体肾炎的发病过程中发挥重要的作用。
Objective To investigate the expression and role of CD44 and its ligand osteopontin (OPN) in the development of experimental crescentic glomerulonephritis. Methods Rabbit anti-nephrotoxicity serum was used to make crescentic glomerulonephritis model. The expressions of CD44, OPN and other markers were detected by immunohistochemical double labeling and in situ hybridization. Results The expressions of CD44 and OPN were significantly increased in the development of crescentic glomerulonephritis, and were closely related to the local infiltration of macrophages in the kidney, the formation of crescent and the impairment of renal function. Conclusion In experimental crescentic glomerulonephritis model, CD44 and OPN are highly expressed in glomerular parenchymal cells and tubulointerstitial cells and may play an important role in the pathogenesis of crescentic glomerulonephritis.
