目的 探讨星形细胞瘤组织中Fas和FasL的表达及其与细胞凋亡的关系。方法 采用免疫组织化学方法和TUNEL技术检测 41例星形细胞瘤组织中Fas和FasL的表达以及肿瘤细胞的凋亡指数。结果 2 7例 ( 66% )星形细胞瘤有Fas的阳性表达 ,Fas的表达水平较低 ,3 4例 ( 83 % )的瘤细胞是Fas的表达完全丢失或阳性细胞数 <5 % ;3 2例 ( 78% )星形细胞瘤有FasL的阳性表达 ,FasL的表达水平较高 ,19例 ( 4 6% )FasL的阳性细胞数较多 (阳性细胞占肿瘤细胞 >5 % )。41例星形细胞瘤凋亡指数为 1.0 9± 0 .48。星形细胞瘤Fas和FasL阳性与否和细胞凋亡指数无明显相关性。结论 Fas和FasL在星形细胞瘤的发生和发展中可能起重要作用 ;星形细胞瘤的发生和发展与细胞凋亡异常有关 ;星形细胞瘤Fas的表达水平低下可能导致肿瘤细胞逃脱机体对其的清除作用 ,Fas系统介导细胞凋亡的机制有待于进一步研究
Objective To investigate the expression of Fas and FasL in astrocytomas and its relationship with apoptosis. Methods The expression of Fas and FasL and the apoptosis index of tumor cells in 41 astrocytoma tissues were detected by immunohistochemistry and TUNEL. Results The positive expression of Fas was found in 27 cases (66%) of astrocytoma, and the expression level of Fas was low. The expression of Fas was completely lost or the number of positive cells was less than 5% in 34 cases (83%);3 Two cases (78%) of astrocytoma had positive expression of FasL, and FasL expression was high. In 19 cases (46%), FasL positive cells were more (positive cells accounted for >5% of tumor cells). The apoptotic index of 41 astrocytomas was 1.09±0.48. There was no significant correlation between apoptotic Fas and FasL and apoptotic index in astrocytomas. Conclusion Fas and FasL may play an important role in the occurrence and development of astrocytoma. The occurrence and development of astrocytoma are related to the abnormal apoptosis. The low expression level of Fas in astrocytoma may lead to the escape of tumor cells. Its scavenging effect, the mechanism of Fas system-mediated apoptosis, needs further study
