目的研究转基因牛乳腺生物反应器表达的重组人乳铁蛋白(rh LF)对子代大鼠免疫功能的影响。方法参照一代繁殖毒性扩展试验指南设计试验,将Wister大鼠分为对照组(酪蛋白)、重组人乳铁蛋白组,每组30只雌鼠、15只雄鼠;按照蛋白占比为20%的比重掺入饲料进行全食物喂养。雌/雄大鼠喂养相应饲料两周后进行交配,各组雌鼠在交配期、妊娠期和哺乳期继续给予相应饲料,子代大鼠从断乳到处死继续给予相应饲料。子代大鼠分别在PND0(出生后0天)、PND21(出生后21天)和PND56(出生后56天)进行免疫功能评价。结果重组人乳铁蛋白组与对照组比较,重组人乳铁蛋白组子代大鼠血常规、血生化和淋巴细胞分型结果未见明显改变;溶血空斑试验(PFC)、淋巴细胞增殖试验及自然杀伤细胞(NK细胞)活性试验指标结果均差异无统计学意义(P>0.05)。结论在本研究的试验条件下,重组人乳铁蛋白对子代大鼠免疫功能未见不良影响。
Objective To investigate the effect of recombinant human lactoferrin (rh LF) expressed on transgenic bovine mammary gland bioreactor on immune function in offspring rats. Methods According to the guideline design of reproductive toxicity expansion test, Wister rats were divided into control group (casein), recombinant human lactoferrin group, 30 female rats and 15 male rats. Results Compared with the control group, no significant changes were found in the blood routine, blood biochemical and lymphocyte typing of the recombinant human lactoferrin group. The hemolytic plaque assay (PFC), lymphocyte proliferation test And natural killer cells (NK cells) activity test results showed no significant difference (P> 0.05). Conclusion Under the experimental conditions of this study, recombinant human lactoferrin had no adverse effects on the immune function of offspring rats.
