为了明确糯质和非糯谷子Waxy基因c DNA序列变异,为品质育种提供实验依据,以2份糯质和2份非糯谷子为材料,利用特异RT-PCR和序列比对,研究了Waxy基因exon4~exon9区间c DNA序列SNPs及其氨基酸多态性,结果表明,糯质红粘谷(00000537)与白米粘谷的同源性为99.12%,在第163、第192、第245和第358位点检测到4个SNPs;非糯类型丰田501与赤谷4号的同源性为98.9%,在第139、第189、第282、第445和第451位点有5个SNPs。糯质红粘谷(00000537)与非糯品种赤谷4号相似性为98.23%,红粘谷(00000537)在第139、163、189、192、245、358、445和451位点碱基分别为T、G、T、G、G、A、C和A,赤谷4号在上述位点依次分别为C、A、G、A、A、C、T和G。糯质红粘谷(00000537)与非糯品种丰田501的相似性为98.9%,红粘谷(00000537)在163、192、245、282和358位点分别为G、G、G、A和A,丰田501依次分别为A、A、A、G和C。红粘谷(00000537)和赤谷4号exon4~exon9区间的c DNA序列编码的氨基酸序列中,检测出6个氨基酸变异,和丰田501出现3个氨基酸的变异。
In order to clarify the mutation of Waxy gene c DNA sequence of waxy and non-waxy millet, we provided experimental evidence for quality breeding. Two waxy and two non-waxy millet were used as materials to study Waxy gene by specific RT-PCR and sequence alignment exon4 ~ exon9 interval c DNA sequence SNPs and their amino acid polymorphisms, the results showed that the waxy red sticky valley (00000537) and white rice sticky valley of 99.12% homology, in 163, 192, 245 and 358 Four SNPs were detected in the locus; 98.9% in non-waxy type Toyota 501 and Red Valley 4, and 5 SNPs in the 139th, 189th, 282th, 445th and 451th loci. The similarities of waxy red glutinous rice (00000537) and non-waxy red rice (Chiyu 4) were 98.23%. The bases of red glutinous rice (00000537) at the positions 139,163,189,192,245,358,445 and 451 were T, G, T, G, G, A, C and A. The Red Valley 4 is C, A, G, A, A, C, T and G at the above sites respectively. The similarity of waxy red glutinous rice (00000537) to non-waxy variety Toyota 501 was 98.9%, while that of red glutinous rice (00000537) was G, G, G, A and A at 163,192,245,282 and 358, respectively Toyota 501 followed by A, A, A, G and C, respectively. Six amino acid variations were detected in the amino acid sequence encoded by the c DNA sequence of Hongnong Valley (00000537) and the exon4 ~ exon9 region of Chichi 4, and a variation of 3 amino acids in Toyota 501 was observed.
