目的:观察复方通络胶囊对大脑中动脉缺血再灌注(MCAO)大鼠的预防性脑保护作用。方法:采用MCAO大鼠模型,造模前大鼠灌胃给药1周,以血浆血管性血友病因子(vWF)、血栓调节蛋白(TM)含量和基质金属蛋白酶-2(MMP-2)mRNA的表达为指标,观察复方通络胶囊对MCAO大鼠的作用机制。结果:复方通络胶囊能明显降低模型动物血浆vWF、TM含量,与模型组比较差异显著(P<0.05),并且能抑制MMP-2 mRNA的表达。结论:复方通络胶囊能够降低模型动物血浆vWF、TM含量,抑制MMP-2 mRNA的表达,对MCAO大鼠有预防性脑保护作用。
Objective: To observe the prophylactic brain protective effect of Fufang Tongluo Capsule on middle cerebral artery occlusion (MCAO) in rats. Methods: MCAO rat model was established. The rats were given intragastric administration of vWF, TM and MMP-2 for one week. mRNA expression as an indicator to observe the mechanism of compound Tongluo capsule on MCAO rats. Results: Fufang Tongluo Capsule could significantly reduce vWF and TM content in model animals, which was significantly different from that in model group (P <0.05), and could inhibit the expression of MMP-2 mRNA. Conclusion: Fufang Tongluo Capsule can reduce vWF, TM content in plasma and inhibit the expression of MMP-2 mRNA in the model animal, and can prevent cerebral ischemia in MCAO rats.
