血清和糖皮质激素调节蛋白激酶(SGK)是一个与蛋白激酶B(PKB/AKT)等第二信使蛋白具有极高的同源性的丝/苏氨酸(Ser/Thr)蛋白激酶。调节机制与其他蛋白激酶显著不同,SGK除了受磷酸化/去磷酸化调节外,还存在快速转录水平上的调节机制。它可能作为多种细胞信号转导通路和细胞磷酸化级联反应的一个功能性交汇点,参与离子通道调节、细胞增殖、分化、存活及凋亡的信号转导,在中枢神经系统发育、神经元变性、凋亡过程中起重要作用。本文就SGK的结构、功能及其在中枢神经系统疾病中的研究进展作一综述。
Serum and glucocorticoid-regulated protein kinase (SGK) is a serine / threonine (Ser / Thr) protein kinase with very high homology to the second messenger protein such as protein kinase B (PKB / AKT). Regulatory mechanism is significantly different from other protein kinases. In addition to its regulation by phosphorylation / dephosphorylation, SGK also has a regulatory mechanism at the fast transcriptional level. It may serve as a functional crossover point for a variety of cell signaling pathways and cellular phosphorylation cascades and is involved in the regulation of ion channels, signal transduction of cell proliferation, differentiation, survival and apoptosis in the development of central nervous system, Yuan degeneration, apoptosis plays an important role in the process. This review summarizes the research progress of SGK in its structure, function and disease in central nervous system.
