采用Franz扩散池进行实验,以半透膜和小鼠皮肤为透过屏障,采用HPLC测定接受液[PEG400-95%乙醇-水(1∶3∶6)]中丹皮酚的含量,考察活血止痛凝胶体外释放和经皮渗透规律。结果显示,活血止痛凝胶中丹皮酚的释药规律符合Weibull方程,拟合方程为ln[-1/(1-Q)]=-0.790 5lnt-1.701 2(r=0.980 9),在8 h内丹皮酚的累积释放率为85.18%,释放速率为2.827μg·cm-2·h-1;丹皮酚的累积经皮渗透率以零级模型拟合好,拟合方程为Q(t)=1.757 9t+0.721 3(r=0.999 1),给药8 h后丹皮酚的累积经皮渗透率为54.85%,渗透速率为1.820μg·cm-2·h-1。研究结果表明,活血止痛凝胶具有良好的释放透皮性能。
The Franz diffusion cell was used for the experiment. The transmembrane and mouse skin were used as the barriers, and the content of paeonol in the recipient fluid [PEG400-95% ethanol-water (1:3:6)] was determined by HPLC to investigate the blood circulation. The cumulative percutaneous permeability of paeonol was fitted by a zero-order model and the fitting equation was Q ( t)=1.757 9t+0.721 3 (r=0.999 1). After 8 h of administration, the cumulative percutaneous permeability of paeonol was 54.85%, and the permeation rate was 1.820 μg·cm-2·h-1. The results of the study showed that the Huoxue Analgesic Gel has good release transdermal properties.
