为观察经转化生长因子β1(TGF-β1)诱导的新生儿脐带血Treg中Foxp3和IL-10表达特点,探讨TGF-β1在诱导产生的新生儿脐带血Treg中的作用。将脐带血单个核细胞用CD3mAb激活后,加IL-2培养扩增,同时加入不同浓度的TGF-β1干预,扩增第4天分别用流式细胞术和RT-PCR技术检测CD4+CD25+TregFoxp3蛋白和Foxp3 mRNA的表达,用ELISA法检测扩增细胞培养上清液中IL-10水平。结果显示,TGF-β1诱导下脐带血CD4+CD25+TregFoxp3蛋白和Foxp3 mRNA的表达明显增加,培养上清中IL-10含量也明显增加。表明,TGF-β1在体外能够促进脐带血CD4+CD25+Treg的Foxp3的表达和IL-10分泌。
To investigate the expression of Foxp3 and IL-10 in neonatal cord blood Tregs induced by TGF-β1 and explore the role of TGF-β1 in induction of neonatal cord blood Tregs. The cord blood mononuclear cells were activated with CD3 mAb, then cultured in IL-2 medium and added with different concentrations of TGF-β1 for intervention. Flow cytometry and RT-PCR were used to detect the CD4 + CD25 + TregFoxp3 protein and Foxp3 mRNA expression, IL-10 level in the cell culture supernatant was detected by ELISA. The results showed that the expression of CD4 + CD25 + TregFoxp3 and Foxp3 mRNA in umbilical cord blood was significantly increased under the condition of TGF-β1, and the content of IL-10 in the culture supernatant also increased obviously. It is indicated that TGF-β1 can promote Foxp3 expression and IL-10 secretion of cord blood CD4 + CD25 + Tregs in vitro.
