目的探讨姜黄素在体外对雄激素非依赖性前列腺癌细胞PC-3生物活性的影响。方法分别采用细胞增殖与活性检测试剂盒(CCK-8)比色法、流式细胞分析、人工基底膜穿透法分析姜黄素对PC-3细胞增殖、凋亡和侵袭的影响。采用Western blot法检测姜黄素在体外对核因子κB(NF-κB)、蛋白激酶(Akt)、尿激酶型纤溶酶原激活物(uPA)蛋白表达的影响。结果姜黄素干预后,PC-3细胞的增殖水平和细胞迁移率下降,而凋亡率提高(P<0.01)。姜黄素能在体外抑制PC-3细胞的NF-κB、Akt、uPA蛋白的表达(P<0.01)。结论姜黄素可在体外显著抑制雄激素非依赖性前列腺癌细胞PC-3的增殖和侵袭,并诱导细胞凋亡;其机制可能通过抑制NF-κB、Akt、uPA等信号通路。
Objective To investigate the effect of curcumin on the biological activity of androgen-independent prostate cancer PC-3 in vitro. Methods The effects of curcumin on the proliferation, apoptosis and invasion of PC-3 cells were analyzed by CCK-8 assay, flow cytometry analysis and artificial basement membrane penetration assay respectively. Western blot was used to detect the effect of curcumin on the expression of NF-κB, Akt and uPA in vitro. Results After the intervention of curcumin, the proliferation and cell migration of PC-3 cells decreased and the apoptosis rate increased (P <0.01). Curcumin inhibited the expression of NF-κB, Akt and uPA in PC-3 cells in vitro (P <0.01). CONCLUSION: Curcumin can significantly inhibit the proliferation and invasion of androgen-independent prostate cancer cell line PC-3 in vitro and induce apoptosis. The mechanism may be through the inhibition of NF-κB, Akt, uPA and other signaling pathways.
