与肌浆膜结合的、ATP酶依赖性钙泵活性,仅在一定的实验条件下方可保持。这些实验条件是:(1)温和的匀浆制作方法。采用剪刀力良好的特殊管子,使匀浆操作时间在4℃时大大缩短;(2)在非连续的蔗糖密度梯度范围内分离肌浆膜期间,有一临界性钙浓度(10~(-5)~3×10~(-5)M。浓度高于3×10~(-7)时,可使自肌浆网分离肌浆膜不完全;浓度低于10~(-7)M(通过加入10~(-4)M的EGTA)时,则钙外流系统的功能状态衰减为零,而肌浆膜蓄积钙的能力仍然完整保留;(3)维持等张性:等比重(Isopyconic)离心和/域贮存于高张溶液中均可破坏肌膜结合钙。在等张性条件下离心5分钟,几乎可保留小囊泡状的肌浆膜。

Centrifugation in isotonic conditions for 5 minutes, the vesicle-like sarcoplasmic membrane can be retained almost.
