目的 通过比较氧苯达唑(Oxibendazole, OBZ)与阿苯达唑(Albendazole,ABZ)灌注治疗小白鼠细粒棘球蚴病(Echinococcosis),观察OBZ抗小白鼠体内细粒棘球蚴的药效。 方法 采用随机对照试验方法,将感染细粒棘球蚴 10周的78只小白鼠随机分为5个组:1)阿苯达唑粉剂组(Po ABZ)75.0 mg/kg;2) OBZ 37.5 mg/kg 组;3)OBZ 75.0 mg/kg 组;4)OBZ 150.0 mg/kg 组;5) 生理盐水对照组(NS),每次0.3 ml/只。每周用药3次,连续用药10周后剖检。测定棘球蚴囊湿重和减重率;观察棘球蚴囊的病理形态学改变及超微结构变化,用 HPLC法测定血液、肝脏、肺脏中的药物浓度。 结果 OBZ低剂量(37.5 mg/kg)、中剂量(75.0 mg/kg)和高剂量(150.0 mg/kg)组小鼠棘球蚴囊平均湿重分别为(159.33±263. 21) mg、(49. 75±43. 62) mg 和(191. 5±98. 36) mg,平均减重率分别为 82. 49%、94. 53%和78.96%。ABZ组平均囊湿重和减重率分别为(145.13±162.47) mg和84.05%;各药物治疗组与 NS对照组(910.11±1 496.28) mg比较差异均有显著性(P<0.05);OBZ 75.0 mg/kg、150.0 mg/kg组与ABZ(75.0 mg/kg)比较差异无显著性(P>0.05)。OBZ不同剂量组小鼠囊壁病理改变只数和程度与空白对照组比较差异均有显著性,而与 ABZ组比较差异无显著性。 结论 OBZ具有?
Objective To compare the effects of oxibendazole (OBZ) and albendazole (ABZ) infusion on Echinococcosis in mice and observe the efficacy of OBZ anti-mouse Echinococcus granulosus . Methods A randomized controlled trial was conducted in which 78 mice infected with Echinococcus granulosus for 10 weeks were randomly divided into five groups: 1) Po ABZ 75.0 mg / kg; 2) OBZ 37.5 mg / kg group, 3) OBZ 75.0 mg / kg group, 4) OBZ 150.0 mg / kg group, and 5) saline control group (NS), 0.3 ml each. Medication 3 times a week, continuous medication 10 weeks after necropsy. The hydatid weight and weight loss rate of hydatid cyst were measured. The pathological changes and ultrastructural changes of hydatid cyst were observed. The concentrations of drug in blood, liver and lung were determined by HPLC. Results The average wet weight of hydatid cyst in the OBZ low dose (37.5 mg / kg), middle dose (75.0 mg / kg) and high dose (150.0 mg / kg) groups were (159.33 ± 263.21) mg 49.75 ± 43. 62) mg and (191. 5 ± 98. 36) mg, respectively. The average weight loss rates were 82.49%, 94.53% and 78.96% respectively. The average wet weight and weight loss rate of ABZ group were (145.13 ± 162.47) mg and 84.05%, respectively. There was significant difference between the drug treatment group and the NS control group (910.11 ± 1 496.28) mg (P <0.05) There was no significant difference between 75.0 mg / kg, 150.0 mg / kg and ABZ (75.0 mg / kg) (P> 0.05). There were significant differences in the numbers and the degree of pathological changes of cystic wall in the OBZ different dose groups compared with the blank control group, but no significant difference with the ABZ group.
