目的探讨乙型肝炎病毒(HBV)对高尔基体糖蛋白-73(GP73)表达的影响,并探讨其临床意义。方法采用反转录PCR和Western blot印迹法检测Hep G2和Hep G2.2.15中GP73 mRNA和蛋白的表达;将HBV感染性克隆p HBV1.3转染Hep G2细胞,反转录PCR和Western blot印迹法检测GP73 mRNA和蛋白的表达;酶联免疫吸附法(ELISA)检测对照者、慢性乙型肝炎患者和肝癌患者血清GP73的血清水平。结果 GP73在Hep G2.2.15细胞mRNA和蛋白表达水平较Hep G2细胞高;转染了HBV感染性克隆p HBV1.3的Hep G2细胞GP73 mRNA和蛋白表达较对照细胞高;与对照组相比,CHB组患者和HCC组患者的GP73血清水平显著升高,差异有统计学意义(P<0.05)。结论HBV上调GP73的表达水平,其血清学水平与HBV相关疾病进程相关。
Objective To investigate the effect of hepatitis B virus (HBV) on the expression of Golgi glycoprotein-73 (GP73) and to explore its clinical significance. Methods The expression of GP73 mRNA and protein in Hep G2 and Hep G2.2.15 cells was detected by reverse transcription PCR and Western blotting. Hep G2 cells were transfected with HBV infectious clone p HBV1.3, and reverse transcriptase PCR and Western blot Method was used to detect the expression of GP73 mRNA and protein. Serum levels of GP73 in control, chronic hepatitis B and hepatocellular carcinoma patients were detected by enzyme linked immunosorbent assay (ELISA). Results The mRNA and protein expression of GP73 in Hep G2.2.15 cells were higher than those in Hep G2 cells. The expression of GP73 mRNA and protein in Hep G2 cells transfected with HBV-infected clone p HBV1.3 was higher than that in control cells. Serum levels of GP73 in CHB group and HCC group were significantly increased (P <0.05). Conclusion HBV up-regulates the expression of GP73 and its serological level is related to the progress of HBV-related diseases.
