目的探讨HOXA10基因在正常妇女及不明原因不孕患者子宫内膜中的表达及临床意义。方法选取2010年12月至2011年10月期间在新疆医科大学第一附属医院生殖助孕中心就诊的38例对照组妇女及55例不明原因不孕患者,留取分泌期子宫内膜组织。分别采用原位杂交和实时荧光定量PCR技术检测各组子宫内膜组织中HOXA10mRNA表达水平。结果原位杂交法检测显示,对照组中分泌中晚期子宫内膜间质细胞中HOXA10mRNA表达量高于腺上皮细胞(P<0.05);HOXA10mRNA在对照组分泌期上皮细胞、间质细胞中表达量均明显高于不明原因不孕组(P<0.05)。实时荧光定量PCR技术检测显示,HOXA10mRNA在对照组分泌中晚期表达显著高于分泌早期(P<0.01);不明原因不孕患者HOXA10mRNA分泌中晚期高于分泌早期(P<0.05)。不明原因不孕患者子宫内膜HOXA10mRNA基因表达均显著低于对照组(P<0.01)。结论 HOXA10基因在不明原因不孕子宫内膜组织中低表达,可能是影响子宫内膜容受性导致着床障碍的不孕原因之一。
Objective To investigate the expression and clinical significance of HOXA10 gene in endometrium of normal women and patients with unexplained infertility. Methods From December 2010 to October 2011, 38 control women and 55 unexplained infertility patients who received treatment at the Reproductive and Reproductive and Recuperation Center of the First Affiliated Hospital of Xinjiang Medical University were enrolled in this study. Endometrial tissue was collected during the secretory phase. The levels of HOXA10 mRNA in endometrium of each group were detected by in situ hybridization and real-time fluorescence quantitative PCR respectively. Results In situ hybridization assay showed that the expression level of HOXA10 mRNA in endometrial stromal cells in the control group was higher than that in the glandular epithelial cells (P <0.05). The expression of HOXA10 mRNA in epithelial cells and stromal cells in secretory phase of the control group Were significantly higher than unexplained infertility group (P <0.05). Real-time quantitative PCR showed that HOXA10mRNA expression in the control group was significantly higher than that in the early stage of secretion (P <0.01); HOXA10mRNA secretion in the unexplained infertility group was higher than that in the early stage of secretion (P <0.05). The expression of HOXA10mRNA in endometrium of patients with unexplained infertility was significantly lower than that of the control group (P <0.01). Conclusions The low expression of HOXA10 gene in endometrial tissue of unexplained infertility may be one of the causes of infertility that affects endometrial receptivity.
