利用RAPD分子标记,对我国目前推广杂交种的15个主要亲本自交系进行了类群划分。从105个引物中筛选出对15个不同基因型的玉米自交系扩增产物具有多态性的引物6个,它们是E-17、G-14、F-14、N-18、N-06、G-04,依据6个引物扩增谱带建立0、1型数据,计算15个自交系间遗传距离,并进行聚类分析。结果表明:供试的15个自交系共划分为6个类群:Ⅰ类为中黄204、关17、Mo17,属兰卡斯特血缘;Ⅱ类为D黄212、黄早四、汶黄,属塘四平头血缘;Ⅲ类为B37、B73、B84,属瑞德黄马牙血缘;Ⅳ类为旅9、E28、丹340,属旅大红骨系统;Ⅴ类为5003、478;Ⅵ类为自330。试验结果进一步表明:用RAPD标记进行玉米自交系类群划分与系谱法一致,RAPD在玉米自交系类群划分的应用是可行的
Using RAPD markers, 15 major inbred lines of hybrids in China were divided into four groups. Sixty-five primers were screened from 105 primers for polymorphisms of E-17, G-14, F-14, N-18, N- 06 and G-04. Based on the amplified bands of 6 primers, 0 and 1 data were constructed, and the genetic distances among 15 inbred lines were calculated and clustered. The results showed that the 15 inbred lines tested were divided into six groups: Ⅰ, Zhongqing 204, Guan 17 and Mo17, belonging to the Lancaster blood group; Ⅱ were D Huang 212, Huangzao Si, Wen Wen , Belonging to Tongsan Siping blood; B37, B73 and B84 in Class Ⅲ, belonging to Reebok yellow horse teeth bloodline; Class Ⅳ being Brigade 9, E28 and Dan 340, belonging to Luda Redbone System; Class V being 5003,478; Since 330. The results further indicated that the RAPD marker was used to classify maize inbred lines in accordance with the pedigree method, and the application of RAPD in maize inbred lines was feasible
