目的:观察裸鼠肺组织微环境对人鼻咽癌细胞株增殖的促进作用。方法:将人鼻咽癌细胞克隆株F1在体外与鼠肺块共同孵育,然后将带瘤细胞肺块行皮下移植,并与瘤细胞悬液皮下移植瘤比较,观察各组移植瘤的生长特性;采用结晶紫法和流式细胞术(FCM)观察各组癌细胞体外增殖能力、DNA增殖指数(PI)及S期细胞比值。结果:两组皮下移植瘤的组织学形态无显著差异,肺块组移植瘤生长较快并具有明显侵袭性,各组移植瘤回复培养细胞的增殖A值、PI值和S期细胞比值明显增高,以肺块组瘤细胞变化更明显,它们与移植前体外传代瘤细胞之间的差异有显著性(P<0.01,P<0.05)。结论:鼻咽癌细胞在体外与肺块共同孵育或裸鼠体内移植后均能提高其增殖能力;体外鼻咽癌细胞与肺块共孵育能促进其恶性行为的表达。
Objective: To observe the promotion effect of lung microenvironment on proliferation of human nasopharyngeal carcinoma cell line. Methods: Human nasopharyngeal carcinoma cell clone F1 was co-incubated with mouse lung in vitro. Then the tumor-bearing lungs were subcutaneously transplanted and compared with the subcutaneous tumor of tumor cell suspension to observe the growth characteristics The proliferative ability, DNA proliferation index (PI) and S phase cell ratio were observed by crystal violet method and flow cytometry (FCM). Results: There was no significant difference in histological morphology between the two groups. The xenografts in the lung group grew rapidly and were obviously invasive. The proliferative A value, the PI value and the S phase cell ratio were significantly increased (P <0.01, P <0.05). The difference between the two groups was significant (P <0.01, P <0.05). CONCLUSION: Nasopharyngeal carcinoma cells can enhance their proliferative capacity after co-incubated with lungs or in nude mice in vitro. Co-incubation of nasopharyngeal carcinoma cells and lungs can promote the malignant behavior of NPC cells.
