利用改进的聚丙烯酰胺凝胶电泳技术 ,对生产上应用的 53个玉米杂交种及亲本自交系干种子清蛋白 (白蛋白 Albumin)进行了电泳分析。结果表明 ,改进的聚丙烯酰胺凝胶电泳技术分辨率高、鉴别能力强、准确 ;不仅能鉴别杂交种及亲本自交系 ,对血缘关系较近的材料 ,如同母异父单交种、含有同一血缘的不同衍生系也能区分开。利用该技术对 6个杂交种 10份样品的种子纯度进行了检验 ,同时利用田间种植鉴定法进行了测定 ,二者相关达极显著水平 (r=0 .90 0 7) ,说明改进的聚丙烯酰胺凝胶电泳技术用于玉米种子纯度检验是可靠的。
Using improved polyacrylamide gel electrophoresis, 53 maize hybrids and their parental inbred lines (albumin) were analyzed by electrophoresis. The results showed that the improved polyacrylamide gel electrophoresis technique has the advantages of high resolution, strong discrimination ability and accurate identification of hybrids and parental inbred lines. For the close relatives of the parents, Different derived lines of the same bloodline can also be distinguished. Using this technique, the seed purity of 10 hybrids from 6 hybrids was tested, and the correlation between seed purity of the 6 hybrids was tested using the field planting identification method (r = 0.90 0 7), indicating that the improved polypropylene Amylolyte gel electrophoresis is a reliable method for testing the purity of maize seeds.
