目的 观察土牛膝液对宫颈癌热敏感性的影响。方法 用细胞计数集落形成率 ,了解癌细胞的热敏感性 ,用光镜和流式细胞术观察凋亡细胞 ,免疫组化法观察细胞 HSP70蛋白合成情况。结果 经土牛膝预先处理的细胞增高了对热处理的敏感性 ,癌细胞发生了凋亡 ,细胞膜 HSP70蛋白的表达率下降 ,这些效应均与土牛膝的处理剂量相关。结论 土牛膝液能增强对宫颈癌热敏感性 ,增强这种热敏感性的可能机理是通过诱导细胞凋亡和抑制 HSP70蛋白的合成。
Objective To observe the effect of Tuinaishixiao on cervical cancer susceptibility. Methods The rate of colony formation was counted by cell counting to observe the thermal sensitivity of cancer cells. The apoptotic cells were observed by light microscopy and flow cytometry. The synthesis of HSP70 protein was observed by immunohistochemistry. Results Achyranthes bidentata preconditioned cells increased the sensitivity to heat treatment. The apoptosis of cancer cells occurred, and the expression of HSP70 decreased. These effects were related to the dose of Achyranthes bidentata. Conclusion Achyranthes root can enhance the heat sensitivity to cervical cancer, and the possible mechanism of enhancing this heat sensitivity is through inducing apoptosis and inhibiting the synthesis of HSP70 protein.
