目的研究枸杞多糖(LBP)对人前列腺癌DU-145细胞的抑制作用及其机制。方法用不同浓度的LBP分别对细胞作用后,在显微镜下观察细胞形态学变化,用MTT法测其生长抑制率,原位末端标记法(TUNEL)检测细胞凋亡情况,流式细胞术检测细胞生长周期和凋亡率。结果不同浓度的LBP对人前列腺癌DU-145细胞均有抑制作用,并呈时间和剂量依赖性;细胞阻滞于G0/G1期,随着LBP浓度的增加,凋亡细胞增加,呈现一定的剂量-反应关系。结论 LBP对人前列腺癌DU-145细胞增殖有抑制作用,其机制可能与其阻断细胞生长周期,诱导细胞凋亡有关。
Objective To study the inhibitory effect of LBP on human prostate cancer DU-145 cells and its mechanism. Methods The cells were treated with different concentrations of LBP respectively. Morphological changes of the cells were observed under the microscope. The growth inhibition rate was measured by MTT assay. The apoptosis was detected by TUNEL and flow cytometry Growth cycle and apoptosis rate. Results Different concentrations of LBP could inhibit the proliferation of human prostate cancer DU-145 cells in a dose- and time-dependent manner. The cells were arrested at G0 / G1 phase. With the increase of LBP concentration, the number of apoptotic cells increased, Dose-response relationship. Conclusion LBP can inhibit the proliferation of human prostate cancer DU-145 cells, and its mechanism may be related to blocking the cell growth cycle and inducing cell apoptosis.
