目的分离抗人卵巢癌单克隆抗体COC183-B2轻链可变区(VL)基因并测定其核苷酸序列。方法用PCR方法体外扩增单抗COC183-B2VL基因,将其克隆入PUC19载体,重组子用Sanger’s双脱氧链终止法测定其核苷酸序列,将其序列通过Internet与GeneBank、EMBL、DDBJ和PDB已发表的抗体序列进行比较分析。结果VL基因全长336bp,属小鼠免疫球蛋白轻链第Ⅱ亚类(SubgroupⅡ),由种系基因的VK及J基因的MUSJK2重排而成。该VL基因序列已被GeneBank收录(acesionNo.gbAF044077)。结论该VL基因为抗人卵巢癌单抗COC183-B2功能性轻链可变区基因。
Objective To isolate the anti-human ovarian cancer monoclonal antibody COC183-B2 light chain variable region (VL) gene and determine its nucleotide sequence. Methods The monoclonal antibody COC183-B2 VL gene was amplified by PCR in vitro and cloned into the PUC19 vector. The nucleotide sequence of the recombinant was determined by Sanger’s dideoxy chain termination method. The sequence was passed through the Internet and GeneBank, EMBL, and DDBJ. Comparing with published PDB antibody sequences. Results The VL gene was 336 bp in length, belonging to subgroup II of mouse immunoglobulin light chain. It was rearranged from the VK of germline gene and MUSJK2 of J gene. The VL gene sequence has been included in GeneBank (acesionNo.gbAF044077). Conclusion The VL gene is a gene encoding a functional light chain variable region of COC183-B2 in human ovarian cancer.
