用CFU-GM半固体琼脂培养技术研究了抗人胸腺细胞球蛋白(ATG)和抗CD3单克隆抗体(McAb)对18例再生障碍性贫血(AA)患者骨髓CFU-GM体外生长的影响。结果表明,1.加入16.7μg/mlATG或1:50抗CD3McAb,体外均可增加AA患者骨髓CFU-GM的集落数(P<0.05),两者的作用无显著差异(P>0.05)。2.ATG和抗CD3McAb对AA患者干细胞缺乏型和免疫介导型的作用无显著差异(P>0.05)。用此项技术对AA患者作药敏试验,并加测外周血CD8+、DR+细胞比值,可作为选择免疫抑制疗法的参考。
The effects of anti-human thymocyte globulin (ATG) and anti-CD3 monoclonal antibody (McAb) on the growth of bone marrow CFU-GM in 18 patients with aplastic anemia (AA) were studied by CFU-GM semi-solid agar culture. The result shows that: 1. The addition of 16.7μg / ml ATG or 1:50 anti-CD3McAb could increase the number of CFU-GM colonies in AA patients (P <0.05) in vitro. There was no significant difference between the two groups (P> 0.05). 2. There was no significant difference between ATG and anti-CD3McAb on AA patients with SLE and immune-mediated (P> 0.05). With this technology in patients with AA susceptibility testing, and increase the ratio of peripheral blood CD8 +, DR + cells can be used as a reference for the selection of immunosuppressive therapy.
