目的:研究地黄饮子对快速老化小鼠P8线粒体通路细胞凋亡的影响,探讨其治疗AD的药效及作用机制。方法:运用RT-PCR检测地黄饮子对SAMP8小鼠Caspase-3、Caspase-9、Apaf-1基因mRNA表达的影响,观察小鼠细胞凋亡情况。结果:地黄饮子能下调海马Caspase-3mRNA、Caspase-9mRNA、Apaf-1mRNA的表达,且均有统计学意义。结论:地黄饮子可通过对细胞凋亡的线粒体途径的影响,抑制促凋亡基因,减少神经元的丢失,从而改善学习记忆能力。
Objective: To study the effect of Rehmanniae Decoction on the apoptosis of P8 mitochondria pathway in the rapidly aged mice, and to explore its pharmacodynamics and mechanism of action in treating AD. Methods: RT-PCR was used to detect the effect of Rehmanniae Decoction on the mRNA expression of Caspase-3, Caspase-9 and Apaf-1 in SAMP8 mice and the apoptosis of mice was observed. RESULT: Rehmanniae Decoction can down-regulate the expression of Caspase-3mRNA, Caspase-9mRNA and Apaf-1mRNA in hippocampus, all of which have statistical significance. Conclusion: Rehmanniae Decoction can improve the ability of learning and memory by affecting the mitochondrial pathway of apoptosis, inhibiting the pro-apoptotic genes and reducing the loss of neurons.
