目的:对不同产地芡实药材的水溶性蛋白质含量进行分析。方法:以低温研磨提取芡实药材的水溶性蛋白质,以牛血清白蛋白为对照,用考马斯亮蓝G-250显色,采用可见分光光度法测定不同产地芡实药材中水溶性蛋白质含量,并应用聚类分析法对结果进行分析。结果:牛血清白蛋白质量浓度在0.005 01~0.150 3 mg/ml范围内与其吸光度值呈良好的线性关系(r=0.999 2);精密度、稳定性、重复性试验的RSD≤2.04%;平均加样回收率为98.95%,RSD为1.76%(n=6)。样品中水溶性蛋白质的含量平均值为0.457 7 mg/g。聚类分析结果显示,长江流域的芡实水溶性蛋白质含量较高,南方芡实次之,而北方芡实的水溶性蛋白质含量最少。结论:该方法简便可靠、重复性好,适用于芡实药材中水溶性蛋白质的含量测定。
Objective: To analyze the water-soluble protein content of Radix Astragali in different areas. Methods: Water-soluble protein was extracted from Gorgon fruit at low temperature. Bovine serum albumin (BSA) was used as control. Coomassie Brilliant Blue G-250 was used for color development. The content of water-soluble protein was determined by visible spectrophotometry. Analysis of the results of class analysis. Results: The concentration of bovine serum albumin showed a good linear relationship with absorbance value (r = 0.999 2) in the range of 0.005 01 ~ 0.150 3 mg / ml; the RSD of precision, stability and repeatability test was ≤ 2.04%; mean The recovery was 98.95% with a RSD of 1.76% (n = 6). The average content of water-soluble protein in the sample was 0.457 7 mg / g. The results of cluster analysis showed that the content of water-soluble protein was higher in the Yangtze River Basin than that in the south, while the water-soluble protein content in the fruit of the north was the least. Conclusion: The method is simple, reliable and reproducible. It is suitable for the determination of water-soluble protein in Gushi Fructus.
