目的:了解RNA干扰沉默STAT3基因对结肠癌细胞侵袭的可能作用。方法:根据STAT3基因特点,设计合成STAT3小干扰RNA(small interfering RNA,siRNA),并转染人结肠癌细胞系HCT116后,采用RT-PCR检测STAT3mRNA,采用软琼脂集落培养试验检测癌细胞锚着不依赖性增殖,采用Boyden小室模型试验检测癌细胞的侵袭能力。结果:STAT3 siRNA可有效抑制结肠癌细胞集落生长和侵袭能力,且与浓度相关。结论:STAT3 siRNA可抑制结肠癌细胞的侵袭。
Objective: To investigate the possible role of STAT3 gene silencing RNAi in colon cancer cell invasion. METHODS: STAT3 mRNA was designed and synthesized according to the characteristics of STAT3 gene. After transfected into human colon cancer cell line HCT116, STAT3 mRNA was detected by RT-PCR. The anchorage of cancer cells was detected by soft agar colony culture Independent of proliferation, Boyden chamber assay was used to detect invasion of cancer cells. Results: STAT3 siRNA effectively inhibited colony growth and invasion of colon cancer cells, and correlated with the concentration. Conclusion: STAT3 siRNA can inhibit the invasion of colon cancer cells.
