目的:筛选出卷叶贝母鳞茎再生的最佳离体培养条件。方法:通过对卷叶贝母开花期鳞茎的离体培养,研究了不同光照条件、培养温度、激素配比以及病毒唑用量对卷叶贝母鳞茎再生的影响。结果与结论:卷叶贝母鳞茎外植体进行适当的低温暗处理后,再转入光下培养,可明显地减轻外植体的褐化程度;鳞茎再生的最适培养温度为(20±2)℃,激素对鳞茎再生有明显的促进作用,其中以6-BA 2.0 mg/L和NAA 0.2 mg/L组合为最佳;浓度为10 mg/L的病毒唑最适于鳞茎的再生。
OBJECTIVE: To screen out the best conditions of in vitro culture of Bulbus Fritillaria bulbiferatum. Methods: The effects of different light conditions, culture temperature, hormone ratio and the amount of ribavirin on bulb regeneration of Fritillaria cirrhosa were studied in vitro. RESULTS AND CONCLUSION: Bulbophyllum bulbs explants were appropriately dark-treated at low temperature and then transferred to light for culture to reduce the degree of browning of explants. The optimum temperature for bulb regeneration was (20 ± 2) ℃. Hormones promoted the regeneration of bulblets. Among them, 6-BA 2.0 mg / L and NAA 0.2 mg / L were the best combinations. Ribavirin at a concentration of 10 mg / L was the most suitable for bulb regeneration.
