目的:探讨人脑胶质瘤中多种基因蛋白的异常表达及意义。方法:应用免疫组化技术检测102例手术切除人脑胶质瘤组织标本中增殖细胞核抗原(PCNA)及p53,p21ras及EGFR蛋白的表达。胶质瘤组织PCNA标记指数(PCNALI)为556±155,p53,p21ras及EGFR蛋白的表达率分别为586%,523%,42%,均显著高于正常脑组织。PCNALI及EGFR蛋白表达与胶质瘤恶性程度相关(P<001),p53,p21ras及EGFR蛋白的表达与胶质瘤恶性程度无关(P>005)。胶质瘤中PCNALI与EGFR蛋白表达无明显相关(P>005),而与p53和p21ras表达相关(P<005)。结论:p53,p21ras和EGFR异常表达与胶质瘤的发生发展相关,PCNALI是评估增殖活性的良好指标。
Objective: To investigate the abnormal expression of multiple gene proteins in human gliomas and its significance. Methods: The expression of proliferating cell nuclear antigen (PCNA), p53, p21ras and EGFR protein in 102 cases of resected human glioma specimens were detected by immunohistochemistry. The PCNA labeling index (PCNAALI) of glioma was 556 ± 155, the expression rates of p53, p21ras and EGFR were 586%, 523% and 42% respectively, which were significantly higher than those in normal brain tissue. The expressions of PCNALI and EGFR were correlated with the malignant degree of glioma (P <001). The expressions of p53, p21ras and EGFR were not associated with the malignant degree of glioma (P> 0.05). There was no significant correlation between PCNALI expression and EGFR protein expression in gliomas (P> 0.05), but it was correlated with p53 and p21ras expression (P <005). Conclusion: The abnormal expression of p53, p21ras and EGFR is related to the development of glioma. PCNALI is a good indicator to evaluate the proliferative activity.
