为研究水稻MADS盒基因与形态发生的关系,采用一个MADS盒基因家族保守区域特异的引物,运用RT-PCR方法,从水稻珍汕97B的幼穗中分离和克隆了两个新的MADS盒cDNA,命名为nmads1和nmads3。基因序列分析表明,nmads1和nmads3都含有植物MADS盒基因典型的MIK区结构,其中nmads1属于MADS盒基因的GLO亚家族,nmads3属于AGL2亚家族。分子杂交实验发现,nmads1和nmads3在水稻愈伤组织分化期和幼穗期活跃表达,在营养生长阶段的苗期表达受抑制,并发现处在同一发育阶段的水稻幼穗中,核质互作雄性不育系珍汕97A比其保持系珍汕97B多出了一个特异表达的nmads1的基因产物。
In order to study the relationship between the MADS cassette gene and morphogenesis in rice, two new MADS cassette cDNAs were isolated and cloned from the young panicle of rice Zhenshan 97B using a conservative region-specific primer of the MADS box gene family using RT-PCR , Named nmads1 and nmads3. Sequence analysis showed that both nmads1 and nmads3 contained the typical MIK region of plant MADS box gene, in which nmads1 belonged to GLO subfamily of MADS box gene and nmads3 belonged to AGL2 subfamily. The results of molecular hybridization showed that nmads1 and nmads3 were highly expressed in the callus differentiation stage and young panicle stage of rice and inhibited in seedling stage of vegetative growth stage. The male sterile line Zhenshan 97A outperformed its maintainer line Zhenshan 97B with a gene product that specifically expressed nmads1.
