目的:研究小檗碱和环孢素联用时大鼠肝组织中肿瘤及免疫相关基因表达谱的影响。方法:大鼠予以盐酸小檗碱ig 6 d,最后一天予以环孢素单次ig给药。取肝脏提取总RNA。用Cy3和Cy5两种荧光物质分别标记阴性时照组,环孢素组和环孢素+盐酸小檗碱组肝脏mRNA,制备成探针与表达谱芯片进行杂交。结果:在被检测的4 096个基因中,环孢素组与肿瘤及免疫相关的差异表达基因共3个,均为表达下调。环孢素+盐酸小檗碱组与肿瘤及免疫相关的盖异表达基因共14个,其中表达上调的相关基因5个,表达下调的相关基因9个。结论:小檗碱和环孢素联用可影响大鼠肝组织中肿瘤及免疫相关基因的表达。
OBJECTIVE: To study the effects of berberine and cyclosporin on the expression of tumor and immune related genes in rat liver tissue. Methods: Rats were given berberine hydrochloride ig for 6 days, and the last day was given cyclosporine single ig. Take the liver to extract total RNA. Cy3 and Cy5 were used to label the negative control group, cyclosporine group and ciclosporin + berberine hydrochloride group, respectively, to prepare probes for hybridization with the expression profiling chip. Results: Of the 4 096 genes examined, 3 genes were differentially expressed in the ciclosporin group, both of which were downregulated. Ciclosporin + berberine hydrochloride group and cancer and immune-related gene expression of the cover a total of 14, of which 5 were up-regulated genes, down-regulated 9 genes. Conclusion: Combination of berberine and cyclosporine can affect the expression of tumor and immune related genes in rat liver tissue.
