为了探讨脑缺血再灌注(CIR)大鼠额叶皮质细胞内Bcl-2和HSP70的表达及川芎嗪、尼莫地平的干预作用,本研究采用Bannister’s颈动脉血引流法复制Wistar大鼠CIR模型,将动物随机分成6组,即正常对照组、假手术组、模型组、川芎嗪治疗组、尼莫地平治疗组和川芎嗪+尼莫地平联合治疗组,每组各15只动物。川芎嗪治疗组按100mg/kg经腹腔注射,尼莫地平组经静脉给药(1mg/kg),联合治疗组则同时给上述剂量药物。三个治疗组均自缺血起给药一次,然后每24h给药一次。所有动物额叶皮质的切片均采用免疫组织化学技术检测Bcl-2和HSP70阳性细胞的表达情况。结果显示:正常对照组Bcl-2和HSP70无阳性表达,假手术组仅HSP70有少量表达,模型组Bcl-2和HSP70阳性细胞数明显有所增多,而川芎嗪或尼莫地平治疗组Bcl-2和HSP70阳性细胞数明显多于模型组(P<0.05)。与单独用药组相比,川芎嗪+尼莫地平联合治疗组Bcl-2、HSP70阳性细胞数明显增多(P<0.05)。以上结果表明川芎秦和尼莫地平能够上调CIR时Bcl-2和HSP70的表达,提示它们可能在CIR中发挥保护作用。
To investigate the effects of ligustrazine and nimodipine on the expression of Bcl-2 and HSP70 in frontal cortex of rats with cerebral ischemia and reperfusion (CIR) and the effect of ligustrazine and nimodipine on the CIR model of Wistar rats The animals were randomly divided into 6 groups: normal control group, sham operation group, model group, ligustrazine treatment group, nimodipine treatment group and ligustrazine + nimodipine combined treatment group with 15 animals in each group. Ligustrazine treatment group by 100mg / kg by intraperitoneal injection, nimodipine group by intravenous administration (1mg / kg), combined with the treatment group while giving the above dose of drug. Each of the three treatment groups was given once from ischemia and then every 24 hours. Immunohistochemistry was used to detect the expression of Bcl-2 and HSP70-positive cells in all animal frontal cortex sections. The results showed that there was no positive expression of Bcl-2 and HSP70 in normal control group, only a small amount of HSP70 was detected in sham operation group, the number of Bcl-2 and HSP70 positive cells in model group was significantly increased, while Bcl- 2 and HSP70 positive cells were significantly more than the model group (P <0.05). Compared with the drug alone group, the Bcl-2 and HSP70 positive cells in the combination of ligustrazine + nimodipine group were significantly increased (P <0.05). The above results show that Chuanxiong Qin and nimodipine can up-regulate the expression of Bcl-2 and HSP70 during CIR, suggesting that they may play a protective role in CIR.
