实验观察了HL-60细胞的一些生物学特点及TPA和RA的分化诱导作用。HL-60细胞在液体培养中呈单个细胞悬浮生长,倍增时间25~31h,饱和密度20×10~5/ml左右。HL-60细胞形态类似早幼粒细胞,具有早幼粒细胞的细胞化学特征,NBT还原反应阴性或弱阳性。HL-60细胞在软琼脂培养中能自发形成集落。TPA诱导HL-60分化为巨噬样细胞。TPA与RA的分化诱导作用均与药物浓度有依赖关系,有效浓度分别为0.162~162nM和1~1000nM。TPA与RA诱导HL-60细胞分化的同时,对细胞增殖有抑制作用。
Some biological characteristics of HL-60 cells and differentiation induction of TPA and RA were observed experimentally. HL-60 cells in liquid culture was a single cell suspension growth, doubling time 25 ~ 31h, saturation density of 20 × 10 ~ 5 / ml or so. HL-60 cells are similar in shape to promyelocytic cells, have cytochemical characteristics of promyelocytic cells, and NBT reduction reaction is negative or weakly positive. HL-60 cells spontaneously formed colonies in soft agar culture. TPA induces the differentiation of HL-60 into macrophage-like cells. The induction of differentiation of TPA and RA is dependent on the drug concentration, with effective concentrations ranging from 0.162 to 162 nM and 1 to 1000 nM, respectively. TPA and RA induced differentiation of HL-60 cells while inhibiting cell proliferation.
