目的讨论干扰素的抗胶原蛋白合成作用的机制.方法以含小鼠胶原α2(Ⅰ)基因上游04kb,08kb,20kb启动片段为研究靶序列,应用CAT作报告基因,结合基因转染技术讨论IFNα,IFNγ对胶原基因的启动抑制活性.结果IFNα,IFNγ单独应用对小鼠胶原α2(Ⅰ)基因上游不同长短启动序列转录活性无阻断作用,但IFNα,IFNγ与TNFα联合应用则具有较强的抑制活性.结论IFNα,IFNγ的抗纤维化活性与其对小鼠α2(Ⅰ)胶原-2kb~+54bp启动活性的作用无直接相关,但IFNα,IFNγ可协助TNFα的抑制活性.
Objective To discuss the mechanism of anti-collagen synthesis of interferon. Methods The promoter sequence of 0 4kb, 08kb and 2 0kb upstream of α2 (Ⅰ) gene of mouse collagen was used as the target sequence. Using CAT as reporter gene and gene transfection technique, the effects of IFNα and IFNγ on the initiation of collagen gene Inhibit activity Results IFNα and IFNγ alone had no effect on the transcriptional activities of collagen α2 (Ⅰ) upstream of mice. However, the combination of IFNα, IFNγ and TNFα had a strong inhibitory activity. Conclusion The anti-fibrotic activity of IFNα and IFNγ is not directly related to their effect on the promoter activity of -2kb ~ + 54bp of α2 (Ⅰ) collagen in mice, but IFNα and IFNγ may contribute to the inhibitory activity of TNFα.
