目的:观察洁泽2号(JZ2)中药复方洁泽1号(JZ1)与外用杀精剂壬苯醇醚(N-9)混合制剂体外对白色念珠菌的抑菌活性及对小鼠白色念珠菌性阴道炎的预防作用。方法:采用琼脂稀释多点接触法检测最小抑菌浓度(MIC)和最小杀菌浓度(MBC);造模前阴道内按分组分别注入各药物凝胶20μl,1min后阴道接种白色念珠菌液5×104个/只,分别于造模后第2、4、7、14天观察小鼠阴道灌洗液菌丝生成及菌落计数结果,阴道、子宫组织PAS染色,各组阴道组织的炎症分值及阴道炎预防率。结果:体外实验中,JZ2对念珠菌的抑制作用较N-9好,菌液浓度为5×105个/ml时,JZ2的MIC和MBC均为1∶64(含JZ17.8mg,N-90.78mg);随着JZ2凝胶中JZ1浓度的降低,小鼠白色念珠菌性阴道炎预防率逐渐下降,呈现一定的量效关系。JZ2高剂量凝胶组预防率可达65.0%,与模型组预防率0、空白凝胶组预防率20.0%比较差异有统计学意义(P<0.01),与西药对照组(联苯苄唑凝胶)作用相当,但强于N-9组,且差异有统计学意义(P<0.05)。JZ2中、低剂量凝胶组预防率较低,但与模型组之间的差异有统计学意义(P<0.05)。结论:体外实验中JZ2(含JZ17.8mg,N-90.78mg)即可抑制念珠菌的生长;JZ2凝胶对白色念珠菌性阴道炎具有较好的预防作用。
OBJECTIVE: To observe the antibacterial activity against Candida albicans in vitro of JZ2 combined with non-phenyl alcohol ether (N-9) Prevention of bacterial vaginosis. Methods: The minimal inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined by agar dilution multiple contact method. 20μl of each drug gel was injected into the vagina by intragastric injection separately. After 1 minute, Candida albicans fluid was inoculated vaginally 5 × 104 / only, were observed on the 2nd, 4th, 7th, and 14th days after modeling respectively, the mycelial growth and colony counting results of vaginal lavage fluid in mice, the vaginal and uterine tissue PAS staining, Vaginitis prevention rate. Results: The inhibitory effect of JZ2 on Candida was better than that of N-9 in in vitro experiments. The MIC and MBC of JZ2 were 1:64 (containing JZ17.8mg and N-90.78 mg). With the decrease of JZ1 concentration in JZ2 gel, the prevention rate of Candida albicans vaginitis in mice gradually decreased, showing a certain dose-effect relationship. The prevention rate of JZ2 high-dose gel group was 65.0%, which was significantly lower than that of the control group (0%) and the control group of blank gel group (20.0%) (P <0.01) Glue), but stronger than the N-9 group, and the difference was statistically significant (P <0.05). In JZ2 medium and low dose gel groups, the prevention rate was lower, but there was a significant difference with model group (P <0.05). Conclusion: JZ2 (containing JZ17.8mg and N-90.78mg) can inhibit the growth of Candida albicans in vitro; JZ2 gel has a good preventive effect against Candida albicans vaginitis.
