目的 评估各种药物对口腔厌氧菌内毒素的降解作用。方法 采用热酚水法及苯酚氯仿石油醚法提取纯化的牙龈卟啉菌、脆弱类杆菌及核梭杆菌内毒素(200μg/m l),分别加入不同浓度药物2m l,在体外37℃孵育15m in、30m in,透析后用鲎试验的偶氮显色法定量检测内毒素的降解程度。结果 降解内毒素的大小次序为:30% 双氧水、50% 枸橼酸、大蒜汁、大蒜1:1 稀释液、25% 枸橼酸、3% 双氧水,它们的作用有浓度依赖性,除双氧水外还有时间依赖性,而溶菌酶的作用极微。结论 提示各种药物降解内毒素的大小和机制不同。
Objective To evaluate the degradation of endotoxin in oral anaerobic bacteria by various drugs. Methods Purified Porphyromonas gingivalis, Bacteroides fragilis and Fusobacterium nucleatum endotoxin (200μg / ml) were extracted by hot phenolic water method and phenolchloroform petroleum ether method and were respectively treated with different concentrations of drugs (2ml) Incubation 15m in, 30m in, after dialysis with arsenic test colorimetric detection of endotoxin degradation quantitatively. Results The order of the degradation of endotoxin was: 30% hydrogen peroxide, 50% citric acid, garlic juice, garlic 1: 1 dilution, 25% citric acid, 3% hydrogen peroxide. There is also time dependence, while the role of lysozyme minimal. Conclusions suggest that various drugs degrade endotoxin in different size and mechanism.
