[目的]研究TLR激动剂LPS和PGN影响中性粒细胞凋亡及凋亡相关的Bax蛋白的表达。[方法]分离培养正常受试者的外周血中性粒细胞,分别加入LPS和PGN两种TLR激动剂,并设生理盐水对照,用流式细胞术检测中性粒细胞的凋亡抑制率,通过Westernbloting技术检测中性粒细胞Bax蛋白的表达。[结果]LPS和PGN降低中性粒细胞自发性凋亡的发生率,与自发性凋亡组比较差异有统计学意义(P﹤0.05);接受LPS和PGN刺激的中性粒细胞表达Bax蛋白均低于自发性凋亡组(P﹤0.05)。[结论]在TLR激动剂诱发的中性粒细胞生存效应中,LPS和PGN可能通过降低中性粒细胞内Bax蛋白的表达来延长中性粒细胞的生命期限。
[Objective] To investigate the effects of TLR agonists LPS and PGN on neutrophil apoptosis and apoptosis-related Bax protein expression. [Method] Peripheral blood neutrophils were isolated and cultured in normal subjects. Two kinds of TLR agonists, LPS and PGN, were respectively added. The normal saline control group was given. The apoptosis inhibition rate of neutrophils was detected by flow cytometry. Neutrophil Bax protein expression was detected by Western blotting. [Results] LPS and PGN decreased the incidence of spontaneous apoptosis of neutrophils, compared with that of spontaneous apoptosis group (P <0.05). The expression of Bax protein in neutrophils stimulated by LPS and PGN Were lower than the spontaneous apoptosis group (P <0.05). [Conclusion] LPS and PGN may extend the life span of neutrophils by decreasing the expression of Bax protein in neutrophils in TLR agonist-induced neutrophil survival effect.
