利用陆地棉自交、陆地棉×亚洲棉、陆地棉×瑟伯氏棉进行了胚珠培养的研究。遗传材料在生长速度、成胚率方面对培养基具有特异性;以陆地棉×亚洲棉为材料研究了生长素和氨基酸对胚生长的影响。IAA 0.5-1ppm和脯氨酸0.1mM分别有效地提高了成胚率。NAA 0.5ppm和谷氨酰胺0.1mM分别使正常胚的比例显著提高;胚珠培养会产生一些不正常胚,有些胚不能培养成苗;使用活性碳能有效地避免幼根发黑现象,促进根的生长和新根的发生;本文对胚珠培养的优越性进行了讨论。
In this study, ovule culture was conducted using upland cotton, cotton × cotton, cotton × Cerberus. The genetic material was specific to culture medium in terms of growth rate and embryogenesis rate. The effects of auxin and amino acids on embryo growth were studied using upland cotton × Asiatic cotton. IAA 0.5-1 ppm and proline 0.1 mM respectively increased the embryogenesis rate. NAA 0.5ppm and glutamine 0.1mM, respectively, so that the proportion of normal embryos significantly increased; ovule culture will produce some abnormal embryos, some embryos can not be cultured into seedlings; the use of activated carbon can effectively prevent young black root phenomenon and promote root Growth and emergence of new roots; the superiority of ovule culture is discussed in this paper.
