目的:观察蚁龙通痹汤对兔膝骨关节炎病理形态及软骨细胞凋亡和凋亡调控基因Bcl-2及其抑制基因Bax的影响,探讨其治疗骨关节炎可能的作用机制。方法:将40只日本雄性大耳白兔按体重分为4组,即正常组,模型组、蚁龙通痹汤组、葡立胶囊组,每组10只。除正常组外,其余各组均行右侧膝关节前交叉韧带切断建立骨关节炎病变模型。造模后第2天开始给药(葡立胶囊0.1 g.kg-1.d-1,蚁龙通痹汤生药0.6 g.kg-1.d-1,溶于10 mL生理盐水中灌胃,每日1次),连续4周。观察各组兔膝关节软骨及滑膜组织病理形态学改变,测定关节液中肿瘤坏死因子(TNF)-α和白介素(IL)-1β含量,并用TUNEL法检测软骨细胞凋亡,分析软骨中凋亡调控基因Bcl-2及其抑制基因Bax表达情况。结果:与正常组比较,模型组动物关节液IL-1β和TNF-α含量、软骨细胞凋亡率、Bcl-2、Bax及Bcl-2/Bax、关节外在表现和软骨病理组织学均出现明显变化;与模型组比较,蚁龙通痹汤组、葡立胶囊组各项指标均有明显改善,差异有统计学意义(P<0.05)。蚁龙通痹汤组、葡立胶囊组组间差异有统计学意义(P<0.05)。结论:蚁龙通痹汤能够减少兔膝骨关节炎关节液IL-1β和TNF-α的分泌,显著降低软骨细胞凋亡率,提高Bcl-2/Bax而达到对软骨组织保护的作用。
OBJECTIVE: To observe the effect of ant dragon Tongbi decoction on pathological changes of osteoarthritis of rabbit knee and the expression of apoptosis-related gene Bcl-2 and its inhibitory gene Bax in rabbit knee, and to explore its possible mechanism of action in the treatment of osteoarthritis. Methods: Forty male Japanese white rabbits were divided into 4 groups according to body weight: normal group, model group, ant dragon Tongbi decoction group and Puli capsule group, with 10 in each group. Except the normal group, all the other groups underwent right anterior cruciate ligament dissection to establish the model of osteoarthritis. On the second day after modeling, the rats were treated with Poria capsule 0.1 g.kg-1.d-1 and ant dragon Tongbi decoction 0.6 g.kg-1.d-1, dissolved in 10 mL of normal saline , Once daily) for 4 weeks. The pathological changes of the articular cartilage and synovial tissue of rabbits in each group were observed. The contents of tumor necrosis factor (TNF) -α and interleukin-1β (IL-1β) in synovial fluid of the rabbits in each group were measured. TUNEL method was used to detect the chondrocyte apoptosis. Expression of Bcl-2 and its suppressor gene Bax. Results: Compared with normal group, IL-1β and TNF-α in synovial fluid, apoptotic rate of chondrocytes, Bcl-2, Bax and Bcl-2 / Bax in joint fluid and cartilage histopathology (P <0.05). Compared with the model group, the indexes of Ailongtongbi Decoction group and Puli capsule group were significantly improved, the difference was statistically significant (P <0.05). There were significant differences between the two groups (P <0.05). CONCLUSION: Ant Longtongbi Decoction can reduce the secretion of IL-1β and TNF-α in articular synovial fluid of rabbit knee osteoarthritis, significantly reduce the apoptosis rate of chondrocytes and increase the Bcl-2 / Bax, which can protect the cartilage tissue.
