目的观察皮肤肌肉切口牵拉术对幼年大鼠机械痛阈和成年后空间学习记忆能力的影响。方法 3周龄幼年大鼠27只,体重60~80 g,随机分成3组(n=9):空白对照组(C组)、假手术组(Sham组)、皮肤/肌肉切口牵拉模型组(skin/muscle incision and retraction,SMIR组)。SMIR组行皮肤肌肉切口牵拉手术,Sham组行肌肉切开未作牵开处理,C组大鼠不进行手术操作。3组大鼠分别在术前、术后1、3、7、12、22、32 d时,测定机械痛阈(MWT)。各时间点测试完毕后,3组大鼠于术后33 d行Morris水迷宫实验。结果 SMIR组大鼠术后1天痛阈降低(P<0.05),3组大鼠在术前及术后3、7、12、22、32 d痛域阈值差异无统计学意义(P>0.05)。Morris水迷宫实验中,与Sham组和C组相比,SMIR组Morris水迷宫定向航行实验平均潜伏期延长(P<0.01);SMIR组大鼠在平台所在象限的游泳时间和路径与总游泳时间和路径的比例显著减小(P<0.01);Sham组与C组间差异均无统计学意义(P>0.05)。结论幼年大鼠SMIR术不引起慢性疼痛,而损害空间学习和记忆功能。
Objective To observe the effect of skin and muscle incision traction on mechanical pain threshold and spatial learning and memory abilities in juvenile rats. Methods 27 young 3-week-old rats weighing 60-80 g were randomly divided into 3 groups (n = 9): blank control group (C group), sham operation group (Sham group), skin / muscle incision traction model group (skin / muscle incision and retraction, SMIR group). SMIR group dermal muscle incision traction surgery, Sham group did not open the line of muscle incision treatment, C group of rats without surgery. The mechanical pain threshold (MWT) was measured in 3 groups at preoperative, 1, 3, 7, 12, 22 and 32 days postoperatively. After testing at all time points, Morris water maze test was performed in 33 rats at 3 days after operation. Results The pain threshold was decreased 1 day after operation in SMIR group (P <0.05). There was no significant difference in the threshold of pain threshold in 3, 12, 22, 32 d before and after operation in the three groups (P> 0.05 ). In the Morris water maze test, the average latency of Morris water maze test in SMIR group was longer than Sham group and C group (P <0.01); the swimming time and path of SMIR rats in the platform quadrant were significantly correlated with the total swimming time and (P <0.01). There was no significant difference between Sham group and C group (P> 0.05). Conclusions SMIR surgery in young rats does not cause chronic pain, impairing spatial learning and memory function.
